Karin Seyfried studierte Ballett an der Wiener Staatsoper und absolvierte ihr Musicaldiplom am Tanz/Gesangsstudio Theater an der Wien. Erste Bühnenerfahrungen sammelte sie parallel zum Studium als Meg in Phantom der Oper, Anne in La Cage aux folles und bei der Uraufführung von Elisabeth. Seit 1995 spielt sie auch an deutschen Bühnen, u.a. die Betty in Sunset Boulevard, Sandy und Rizzo in Grease und Stephanie in Saturday Night Fever. Außerdem stand sie in zwei TV-Filmen und einer Serie (Regie: Peter Weck) vor der Kamera. Für die Uraufführung von Miami Nights wurde sie als beste Hauptdarstellerin in einem Pop/Rock Musical ausgezeichnet. In Stuttgart spielte sie Belle/Babette in Die Schöne und das Biest, Peggy in 42nd Street und Kaiserin Sissi in Elisabeth. Danach folgten Rollen in Jekyll & Hyde, Singing in the rain und Mozart sowie in der Uraufführung der Moulin Rouge Story.
